Synthetic nanodiscs were produced by solubilizing HEK293 cell membrane fractions overexpessing target full-length multi-transmembrane protein with synthetic polymers such as styrene-maleic acid co-polymers (SMAs), forming discoidal nanostructures with target protein intact in lipid bilayer. The nanodiscs were further purified, offering intact membrane proteins in their natural configuration.

Elisa plates were pre-coated with Flag Tag GPR75-Nanodisc (0.2ug/per well). Serial diluted anti-GPR75 monoclonal antibody solutions were added, washed, and incubated with secondary antibody before Elisa reading. Calculated from the data, EC50 for anti-GPR75 monoclonal antibody binding with GPR75-Nanodisc is 303.6ng/ml.
